N2 - 1-acetyl-6, 7-dihydroxyl-1, 2, 3, 4-Tetrahydroisoquinoline(ADTIQ) is a novel endogenous neurotoxins, and related to glucose metabolism. Therefore, ADTIQ may play a key role in increasing the risk for PD in patients with diabetes. But the neurotoxicity of ADTIQ is still unclear. Here, we try to investigate the neurotoxicity of ADTIQ on SH-SY5Y cells. The Annexin V-FITC/PI, MTT, Caspase 3/7 analyses were used to measure the cell viability and apoptosis of ADTIQ, and dopamine transporter (DAT) levels were detected by western blot. The results show that ADTIQ reduced the cell viability and induces apoptosis in SH-SY5Y cells, it also decreases the expression of DAT, suggesting that ADTIQ involved in the pathogenesis of PD. Diabetes induced the increase of ADTIQ, which could be related with Hyperglycemia and death of dopaminergic neurons.
