A Microfluidic Chip for Circulating Tumor Cells RNA Sequencing at Single Cell Level

Abstract

Here we present an integrated microfluidic chip for single cell RNA sequencing of circulating tumor cells (CTCs). All procedures of single cell RNA sequencing of specific cells in human peripheral blood were integrated in a single microfluidic chip, including blood filtration, CTCs enrichment, single CTC isolation and identification, single cell RNA reverse transcription and cDNA amplification. Our results realized the aim of 'whole blood in, SMART-SEQ II product out' at single CTC level in a microfluidic chip.
