Protein profiling of striatum and substantia nigra from hemiparkinsonian rat model by offgel-HPLC-MS/MS

摘要

Parkinson's disease (PD) is a common neurodegenerative disease. The prevalence of PD among people over 65 years old is about 2 % and it is induced by multifactor including age, gene and environment. Recently, proteomics are increasingly being applied to identify biomarkers for disease. In this study, unilateral 6-hydroxydopamine (6-OHDA) in striatum of PD rat model was established first and demonstrated by motor behavior and immunocytochemistry of tyrosine hydroxylase staining. And then offgel as the first dimensional separation method combined with HPLC-MS/MS was used to analyze protein profiling of striatum and substantia nigra from PD rat. In striatum and substantia nigra, there were 858 and 751 proteins identified, respectively and 381 proteins were both in the two regions. GO (Gene Ontology) and KEGG (Kyoto Encyclopedia of Genes and Genomes) annotation revealed that these proteins were related to metabolism, apoptosis and signal transduction. Additionally, several PD related proteins were also involved in this study including ubiquitin carboxyl-terminal hydrolase isozyme L1 (UchI1), heat shock proteins and 14-3-3 proteins. These investigations would provide protein identification basis for PD pathogenesis and be helpful for further study for biomarker discovery.
