Optimization of dynamic immobilization of trypsin onto IAM

Abstract

In this study, Immobilized Artificial Membrane (IAM) was chosen as matrix and trypsin was used as immobilized macromolecule. The effect of different buffer solutions (Borate, Phosphate, and Tris HCl), concentrations, and pH on the immobilizing rate and activity of the immobilized trypsin was investigated. The optimal condition of off-line immobilization was Tris HCl buffer (10mmol/L, pH 8.0). On the basis of the obtained optimal condition, the trypsin enzyme reactor was prepared, and the preliminary result showed that the dynamically immobilized enzyme reactor possessed high activity, which can be specifically inhibited by the trypsin inhibitor.
