N2 - A rat model of Parkinson's disease was established by direct injection of 6-OHDA into the substantia nigra unilaterally. Animals were assessed by D-amphetamine rotation test, anti-TH staining and detection of dopamine and its metabolites by HPLC-ECD. Rotational behavior experiment shown that the achievement ratio of PD model is 48.9%. Anti-TH staining result indicated that 6-OHDA treatment caused extensive loss of TH immunoreactivity in the substantia nigra and their projections to the striatum. Moreover, DA and its metabolites reduced remarkably in the substantia nigra and striatum of 6-OHDA lesioned rat model compared to control. Therefore, by evaluation of experimental animals, unilateral 6-OHDA induced substantia nigra degeneration can establish dependable rat model of Parkinson's disease with higher achievement ratio.
