Activity assay of salsolinol synthase using high performance liquid chromatography-electrochemical detection

Abstract

Salsolinol synthase is an enzyme for synthesizing salsolinol from dopamine and acetaldehyde. It is very important in the metabolic process of catechol isoquinolines such as salsolinol, Ar-methyl-salsolinol. The catechol isoquinolines are closely related to the pathogenesis of Parkinson 's disease ( PD ) , it is supposed that salsolinol synthase may play a significant role in PD. In this paper, the brain of male SD rats was used for isolation. A method for purification and activity detection of salsolinol synthase with excellent sensitivity and stability were developed. The results of ultrafiltration showed that the activity of salsolinol synthase exists in low solute of 3K.
