Nanopost Array Integrated Digital Microfluidics for Exosome Enrichment and Molecular Ionization

摘要

This study presents a nanopost array-embedded digital microfluidic (DMF) platform combined with laser desorption ionization (LDI) mass spectrometry (MS) for the enrichment and direct ionization of exosomes. The nanopost array, surface-modified with functional antibodies, is embedded in the top substrate of the DMF chip, enabling the automated isolation of exosomes from samples in low volumes (<2 μ L). The enriched exosomes on the nanoposts are then subjected to laser pulses, which resonate with the array to produce ionized exosomal molecules. The established platform has achieved a limit of detection (LOD) of 10-7 mol/ L for verapamil. This method allows for direct molecular analysis of DMF-processed samples by MS, offering an efficient and streamlined approach for bioclinical applications, particularly in exosome analysis.
