RP-HPLC determination and pharmacokinetic study of mangiferin in rat plasma after taking traditional chinese medicinal-preparation: Zhimu decoction

Abstract

A sensitive, simple, and accurate method for determination and pharmacokinetic study of mangiferin in rat plasma was developed using RP-HPLC with UV detection. Sample preparations were carried out by protein precipitation with the addition of ocetonitrile, followed by evaporation of the acetonitrile to dryness. The resultant residue was then reconstituted in mobile phase and injected onto a Hypersil C18 analytical column (200 x 4.6 mm, i.d., 5 μm). The mobile phase was acetonitrile-water (12:88, v/v) with 1% glacial acetic acid and 1% tetrahydrofuran. The volidoted LC method developed was used to take a limited view of the pharmacokinetic profile of mongiferin in rat plasma after having taken Zhimu decoction.
