Optimization of the extraction technics for quinatic acid and akeboside Stc from Mutong by orthogonal test

Abstract

Studies the optimization technique in extracting the quinatic acid and akeboside Stc in Mutong from Akebia. Ultrasonic extraction was applied. The variety of solvent, extraction time and reagent proportion were selected as the factors and were numbered in three levels respectively. L9 (34) orthogonal test was adopted. Using the quinatic acid and akeboside Stc as indicator, measuring the content with reversed phase high-performance liquid chromatography (RP-HPLC) and variance analysis as well as directly analyzing on the measurement were carried out. The optimal condition of the orthogonal test was as follows: The solid dissolved in 80% ethanol, ultrasonic extraction lasted for 1.5 hours, and 12.5 times of solvent was needed. The result of methodological investigation for the optimum technique passed the muster. The technique proved to be practical, which laid theoretical foundation for measuring the quinatic acid and akeboside Stc in different kinds of Mutong from Akebia.
